(A) Internal fluorescence ratio in HEK293 cells transfected with WT, R269H, R232C, or R316H construct at rest or stimulated condition (2 μM 4αPDD) is given as basal (left) or peak (right) value (n >3,000 cells for each condition). Mean calcium responses before and during 4αPDD application are given. Significant difference was observed in TRPV4 mutants (R269H, R232C, or R316H) at basal level, and mutants (R269H or R232C) at activated condition when compared to WT-TRPV4 (2-tailed Student t test, *p < 0.005). (B) Application of 4αPDD induced an increase in intracellular calcium ([Ca2+]i). Error bars, mean ± SEM.
