Foxp3 expression in CD4⁺ T cells obtained from mice deficient in l-Selectin, β₇ integrin, or both. Negatively selected CD4⁺ T cells obtained from mice deficient in l-Selectin (CD62L^−/−), β₇ integrin (β₇^−/−), or both (CD62L^−/− × β₇^−/−) were plate into CD3 mAb-coated wells containing IL-2, TGFβ, and RA for 4 days at 37°C. Cells were permeabilized and stained for CD4 and Foxp3. Cells were gated on CD4⁺ T cells and Foxp3 expression was determined by flow cytometry. The shaded curves represent isotype control Ab staining.
