LRH-1 silencing prevents T stimulation of Cyp19 and P450scc expression. Granulosa cells were infected with lentivirus carrying shRNA sequences against luciferase (shLUC), used as control, SF-1 (shSF-1), or LRH-1 (shLRH-1). Total mRNA or nuclear proteins were isolated 96 h after infection. Total mRNA was used for Cyp19 (A), LRH-1 (B), and SF-1 (C) quantification by real-time PCR, whereas nuclear proteins were used for gel shift analysis using the NRHS element as a probe (D). This experiment was performed three times with identical results. Values are the mean ± sem of a single experiment performed in triplicate.
