Selective inhibitors of NADPH oxidase or short interfering RNA (siRNA)-p47phox inhibits glucose-stimulated reactive oxygen species (ROS) generation in insulin-secreting cells. INS 832/13 cells were incubated with either diluent or apocynin (100 μM, 12 h; A) or diphenyleneiodonium chloride (DPI; 5 μM, 2 h; A) or transfected with p47phox siRNA (B and C), following which they were stimulated with either low (2.5 mM; LG) or high glucose (20 mM; HG) for 1 h. ROS generated was quantified as dichlorofluorescein (DCF) fluorescence and expressed as arbitrary units (AU). B: transfection efficiency of p47phox siRNA was determined by immunoblotting.
