HIF regulation of ANK expression. Cells were transfected with A, C and D) HIF-1α-SiRNA (SiHIF-1α) B) HIF-2α-SiRNA (SiHIF-2α) or control siRNA (C) and cultured in hypoxia. Immunofluorescent staining for A) HIF-1α and ANK B) HIF-2α and ANK in HIF silenced cells in hypoxia. Note, the increase in ANK staining in HIF silenced cells when compared with controls. As expected silenced cells show decreased staining for respective HIFs. C) Silencing of HIF-1α results in increased ANK mRNA expression. D) Western blot analysis of ANK in SiHIF-1α transfected cells.
