(A) Strains containing yeast TAF145 (YSW94), TAF90 (LYC-1), TAF68 (YJR18) and TAF47 (YJR11) under the control of the dextrose-repressible GAL1 promoter were grown in galactose to an OD of 0.5–1.0 (GAL), collected, washed and resuspended in dextrose-containing medium. After 10 h in dextrose medium, the cultures were divided into three flasks, and MMS (0.03%) or HU (0.1 M) was added to separate cultures. After an additional 1.5 and 3 h induction period, cells were collected for RNA isolation. Note that the total amount of time in dextrose for these experiments was 11.5 and 13 h, respectively. (B) Western blot. The levels of TAFII (left) and TBP (right) protein were analyzed after 0, 8 and 12 h in dextrose-containing medium. The asterisk on the left of the TAFII145 blot indicates a cross-reacting protein migrating near TAFII145.
