The ability of irradiated pLGECs to stimulate lymphocyte proliferation was suppressed by the expression of IL-10 after transduction. Incorporation of [3H]thymidine by peripheral blood lymphocytes (1 × 105/well) cocultured with AAV-vIL-10-tetON transduced or nontransduced and irradiated pLGECs (1 × 105/well) in an autologous mixed-cell reaction (n = 6). Transduced pLGECs without doxycycline treatment showed increased lymphocytic proliferation compared with the doxycycline-treated group (n = 6). Suppression of lymphocytic proliferation in mixed-cell reaction with transduced pLGECs with doxycycline treatment was neutralized by IL-10 antibody (n = 6). *Statistically significant differences between irradiated nontransduced pLGEsC + PBL and all other groups.
