T. brucei growth is not affected by RNAi depletion of any single PAT mRNA. Tetracycline-inducible PAT RNAi cell lines were cultured at an initial density of 5 × 103 cells/ml and propagated for 10 days in the presence (closed squares) or absence (open circles) of 1 μg/ml tetracycline. Every 48 h, cell density was quantified with an automated cell counter and cells were diluted as necessary to prevent entry into stationary phase.
