N terminus of flagellin can relieve requirement for NAIP5 in flagellin sensing even when expressed on a distinct molecule from the C terminus. A retroviral construct expressing the N-terminal 65 amino acids of flagellin fused to GFP, and a separate retroviral construct expressing the C-terminal 65 amino acids of flagellin fused to mCherry, were transduced simultaneously into wild-type (A), Naip5−/− (B), or Nlrc4−/− (C) macrophages, and the percentage of cells expressing GFP and/or mCherry was enumerated by flow cytometry 4 days posttransduction. The percentage of double-positive cells among all transduced cells is indicated in parentheses. The results are representative of two representative experiments.
