A Cse4-specific antibody brings down an STB reporter plasmid in a Rep1- and Rep2-dependent and Ndc10 independent manner. Plasmid pulldowns in this set of assays and those for which results are shown in subsequent figures were performed as described in Materials and Methods. The antibody employed was directed to the Myc12 epitope fused to Cse4. Plasmid DNA was detected by Southern blot analysis using a radioactively labeled hybridization probe.
