Forced expression of tapasin is not sufficient to rescue MHC I incorporation into the PLC in HCMV-infected cells. Tapasin-transfected or control MRC-5 cells were mock infected or infected with AD169 or with the HB5 (ΔUS2-US6)-derived ΔΔΔFcγR HCMV deletion mutant, as indicated, at an MOI of 5. Cells were labeled for 3 h prior to lysis at 48 h p.i. Immunoprecipitations were performed as indicated. Proteins were separated by 10 to 11.5% SDS-PAGE.
