Down-regulation of SPAG9 expression using plasmid-based siRNA in colon cancer cells in vitro. Two independent siRNA target sequences directed against SPAG9 (designated SPAG9 siRNA-I and SPAG9 siRNA) and control siRNA (scrambled SPAG9) were used. A: COLO 205 and HCT 116 cells treated with SPAG9 siRNA revealed ablation of SPAG9 protein, compared with SPAG9 siRNA-I, by Western blotting analysis. No reduction in SPAG9 protein was detected with control siRNA. B and C: Reduced SPAG9 expression suppressed the cellular proliferation (B) and colony forming ability (C) of COLO 205 and HCT 116 cells. Cells were counted, and the results are representative of three independent experiments performed in triplicate. Data are reported as means ± SE of triplicate plates. *P < 0.0001.
