Hypoxia stimulates proliferation in MKP-1^+/+ and MKP-1^+/− but not in MKP-1^−/− MEFs. In MKP-1^+/+ MEFs, hypoxia-induced replication was attenuated by inhibitors of MAP kinase pathway (n = 8 replicate wells with 3 to 4 populations of MEFs, each isolated and cultured from individual embryo). *P < 0.05 compared with absorbance value of day 0 DMSO for cells from all three genotypes. **P < 0.001 compared with day 3 DMSO under normoxic condition for MKP-1^+/+ and MKP-1^+/− MEFs. ***P < 0.001 compared with day 3 DMSO under hypoxic conditions for all three genotypes. ^†P < 0.001 compared with day 3 DMSO under normoxic conditions for MKP-1^+/− and MKP-1^−/− MEFs.
