Strains: S. pyogenes WT (SF370), Δpre-crRNA, ΔtracrRNA, Δrnc and Δcsn1. Graph bars, mean values of colony forming units (CFU) per μg of plasmid DNA; error bars, standard deviation (SD); n≥3. pEC287 is tolerated by the Δpre-crRNA, ΔtracrRNA, Δrnc and Δcsn1 mutants but not by the WT strain. As control, transformants in all strains were obtained with the backbone plasmid (Supplementary Fig. 9).
