Comparison of adherent cell concentrations (cell #/cm2) derived by four different methods. The original X–Y images of Ppu2440 (miniTn7PA1/03/04egfp-a) biofilm were taken with an epifluorescent microscope equipped with a ×100 oil immersion lens. Methods 1–3 are image-analysis methods that employ subtle, different ways of processing the original adherent cell image. Biofilm cell concentrations were also determined by destructive removal of biofilm and assessment of the suspended cell numbers
