Dynamin-2 siRNA inhibits lamellipodia formation. (A) SNB 19 glioblastoma cells were plated on glass coverslips and treated with two different siRNA oligomers against dynamin 2 (dyn2) or luciferase (control) for 48 h. After serum starvation for 16 h, 52 h after transfection, cells were stimulated for 10 min with 10% FBS. After fixation in 4% formaldehyde, cells were stained with FITC-labeled phalloidin to visualize lamellipodia formation. Bar, 20 μm. (B) Quantitation of lamellipodial extent as in Figure 1J. mean ± SEM; * p < 0.001. (C) SNB 19 cells were transfected with siRNA directed against either dynamin-2 or luciferase and harvested 48 h posttransfection. Expression of dynamin-2 was determined using Western blot with an anti-dynamin-2 antibody. An antibody against cortactin controls for nonspecific alterations in expression of cytoskeletal proteins.
