Southern and western blot analyses of knock-in parasites. (A) Genomic DNA from wild type (WT), Δldodc::NEOldodc::HYGldodc), and the four knock-in clones (clones 1, 3, 4, and 5) was digested with Sal I and hybridized separately to either LdODC, HYG, or NEO as indicated. Molecular weight markers are shown to the left. (B) Cell lysates from wild type (WT), Δldodc::NEOldodc::HYGldodc), and the four knock-in clones were fractionated by SDS polyacrylamide gel electrophoresis and probed with previously characterized rabbit polyclonal antibodies to LdODC and LdSPDSYN (as loading control).
