Recombinant FH5 FH2 and FH5 FH1FH2 Cap Actin Microfilaments.
(A) Kinetics of elongation of the barbed-end of actin filaments in the presence of FH5 FH2. Preformed actin filament seeds (0.8 μM) were incubated with various concentrations of FH5 FH2 (from top to bottom: 0, 2.5, 3, 4, 5, 6, 8, and 10 nM) before addition of 1.0 μM pyrene-actin monomers. a.u., arbitrary units.

(C) and (D) Kinetics of actin filament depolymerization in the presence of various concentrations of FH5 FH1FH2 (C) (from bottom to top: 0, 10, 20, 50, 100, and 200 nM) and FH5 FH2 (D) (from bottom to top: 0, 5, 10, 20, and 50 nM). FH5 FH1FH2 or FH5 FH2 were incubated with 5 μM actin filaments (80% pyrene-labeled) for 5 min before dilution of the solution 25-fold in buffer G.
