Titration of CYP51F1 with azole compounds. Purified CYP51F1 (2 μM) was dissolved in 100 mM potassium phosphate buffer (pH 7.4) and the absorption spectrum of the ferric form (350–500 nm) was recorded with subsequent additions of the azole compound fluconazole, econazole, ketoconazole, or itraconazole. The binding affinities of the azoles calculated from the plots are shown in Table 1.
