The effect of UBR5A depletion on protein synthesis rates is pronounced when cells are grown in rich media. W303-1A and UBHY-R cells were grown in YP galactose, then resuspended either in YP glucose (“Depleted” UBHY-R and “W303-1A/Glucose”) or galactose (“Induced” UBHY-R) as indicated. Duplicate samples were processed independently. [³⁵S]methionine incorporation and protein concentrations were determined as described in Materials and Methods. Protein synthesis rates were measured as cpm incorporated per μg total protein per minute of labeling, then expressed as a percentage of the respective value at time zero.
