Effects of SIRT1 modulators on SAP97 transcription. (A) Effect of the SIRT1 inducer resveratrol on SAP97 mRNA expression. HL-1 cells transfected with HSF-1 were treated with resveratrol (50 µM) for overnight and real-time PCR analysis was performed with SAP97 and β-actin primers. The mean ratio (±SEM) of SAP97 mRNA quantity normalized to β-actin (n = 6 for control and resveratrol groups) is shown. Statistical significance of the differences among each group was tested by Student's t-test: *P < 0.05. (B) Effect of the SIRT1 inhibitor nicotinamide (NAM) on SAP97 mRNA expression. HL-1 cells transfected with HSF-1 were treated with NAM (5 mM) overnight, and real-time PCR (RT-PCR) analysis was performed with SAP97 and β-actin primers.
