The ability of calcitriol to induce hepatic SLCO1B1 mRNA expression in vitro in human Huh6 liver cells and in vivo in mice was assessed by realtime PCR as described in Methods. In each study, expression of SLCO1B1 in vehicle treated controls is set at 1 and relative stimulation induced by calcitriol is shown as fold-change. Cells were treated for 24 hours with 100 nM calcitriol and mice were treated for 4 weeks with 25 ng calcitriol every other day. Induction of 24-hydroxylase (CYP24) was used as a positive control.
