Western blot analysis and graphical representation of BoNT/A mediated cleavage of SNAP-25 in ES cell-derived motoneurons incubated with 1nM BoNT/A in the presence of the indicated inhibitors for 3 hours. Note that while 1 µM TVL only partially blocked SNAP-25 cleavage, BafilomycinA1 completely inhibited BoNT/A mediated SNAP-25 cleavage at any concentration, while the efficacy of the 4A2-4 antibody tightly correlates with the antibody to toxin ratio.
