FIG. 8

Treatment with both TSA and MS-275 causes increased apoptosis. A Dissociated supporting cells treated with both TSA (100 nM) and MS-275 (5 μM) contained increased numbers of pyknotic nuclei (arrow), scale bar 25 μm. B Dissociated cultures were treated for 24 h with either MS-275 (5 μM), TSA (100 μM), or both, and the resulting apoptosis indices (# pyknotic nuclei/# DAPI-stained nuclei per 10,000 μm2) were calculated. Simultaneous treatment with both inhibitors caused a large increase in apoptosis. Immunoreactivity for cleaved caspase 3 (red) was examined in untreated dissociated supporting cells (C), as well as cells treated with either 1 mM VPA for 24 h (D) or 100 nM TSA for 24 h (E). A dramatic increase in cleaved caspase 3 was noted following TSA treatment but not after VPA treatment. Nuclei stained with DAPI (blue). Scale bar 100 μm, error bars: standard deviation, *p < 0.05.
