FT-ICR-MS2 of the two different sizes of ACP active-site-containing tryptic peptides from a sample of holo-LovF not incubated with malonyl-CoA revealed the presence of both a pantetheine ejection ion (Pant) at 261.127 m/z and a phosphopantetheinyl (PPant) ejection ion at 359.104 m/z. The resulting charge-loss parent phosphopeptide was clearly detectable among the fragment ions. (A) MS2 spectrum of the ACP active Y2448-H2538. (B) MS2 spectrum of the ACP active Y2420-H2538.
