Table 4. Relative activity of hNTAN1 mutants.

Reactions of hNTAN1 (4050nM) with 100μM N1-AII were carried out at 37°C in 50mM Tris-HCl, 150mM NaCl, pH 7.5. After 1 min, reactions were quenched and then analyzed by HPLC as described in “Experimental Procedures.”

Enzyme % Relative Activity (v. WT)
WT 100
C160A 100
C75A 0 *
C75S 0
C75T 0
*

Area of the product peak was less than or equal to that observed in an HPLC trace of N1-AII only at the equivalent reaction conditions.
