Silencing MUC1 sensitizes to melphalan- and dexamethasone-induced death. A, The indicated KMS28PE cells were treated with 4 μM melphalan or 50 μM dexamethasone for 72 h, stained with propidium iodide and assayed for sub-G1 DNA content by flow cytometry. The percentage of cells with sub-G1 DNA is indicated in each panel. B, The percentage of apoptotic cells with sub-G1 DNA is expressed as the mean ± SD obtained from three experiments for the indicated control (open bars), melphalan-treated (solid-bars) and dexamethasone-treated (shaded bars) cells. C, The indicated KMS28PE cells were treated with 4 μM melphalan or 50 μM dexamethasone for 24 h and then seeded in 0.4% methycellulose/RPMI-1640 medium with 10% FBS. Colonies were counted at 28 days.
