Physical and functional association of JMJD2B/MLL2 complex with ERα target promoter. (A) Co-occupancy of the TFF1 promoter by JMJD2B and the components of the MLL2 complex. MCF-7 cells were deprived of estrogen for 3 d and then treated with E2 for 1 h. Soluble chromatin was prepared for ChIP and Re-ChIP assays with antibodies against the indicated proteins. (B) The effect of JMJD2B knockdown on H3K9 methylation on TFF1 and EBAG9 promoters. MCF-7 cells were treated with control siRNA or JMJD2B siRNA. Cells were deprived of estrogen for 3 d and then treated with E2 for 1 h. Soluble chromatin was prepared for ChIP assays using the indicated antibodies. GAPDH and U6 were used as negative controls.
