Bacteria were transformed with chromosomal DNA from a CmR derivative of NMB, and cfu counts were determined on selective and non-selective plates. Transformation frequencies were calculated by dividing the cfu numbers of selective plates by the cfu numbers of non-selective plates. The frequency of the mutant was normalized to that of the wild type, which was set as 100%. Data are the means ± standard deviations of two assays done in duplicate. (D) PilQ levels determined by Western blots. Equal amounts of whole cell lysates (8 μg) were resolved on 10% SDS-PAGE gels and transferred to PVDF membranes. The top half of the membrane was probed with PilQ antisera (Tonjum et al., 1998), while the lower half was probed with Opa antisera (Takahashi et al., 2008) for loading controls.
