Man7 isomer analysis. Man7 N-glycans of SDS-PAGE-separated, Protein A-purified scFv-Fcs were released, separated by a capillary porous graphitic carbon column, and detected by LC-ESI-MS. Reference glycans were prepared from P. vulgaris (kidney bean) in a similar manner. In the secretory HA78 constructs, the Man7.1 isoform is predominant, whereas in wt-PhHA78scKDEL and wt-Ph2G12scSEC, the Man7.2 isoform is prevalent.
