Acini were incubated with control and E-subunit specific siRNA in the presence of carbachol (1μM) for 30 min to stimulate uptake. The effects of carbachol were neutralized by atropine treatment (1μM). Acini were further incubated for 2.5 hours to accomplish protein knockdown. Acini were collected by centrifugation solubilized and subjected to western blot analysis. All samples represent the mean +/- SEM of at least 3 experiments. * = p <= 0.05 vs no siRNA. The western blot is representative of one experiment.
