Acini were incubated with control and E-subunit specific siRNA in the presence of carbachol (1μM) for 30 min to stimulate uptake. The effects of carbachol were neutralized by atropine treatment (1μM). Acini were further incubated for 2.5 hours to accomplish protein knockdown. Cerulein was then added to the appropriate wells for 1 hour. Samples were assayed for trypsin and chymotrypsin activity. Results are expressed as fold vs cerulein 100nM. All samples represent the mean +/- SEM of at least 3 experiments. * = p <= 0.05 vs CER 100nM.
