Acini were incubated with control and E-subunit specific siRNA in the presence of carbachol (1μM) for 30 min to stimulate uptake. The effects of carbachol were neutralized by atropine treatment (1μM). Acini were further incubated for 2.5 hours to accomplish protein knockdown. Cerulein was then added to the appropriate wells for 1 hour. Samples were assayed for secreted and total amylase activity; Results are expressed as amylase secretion as a % of total amylase. All samples represent the mean +/- SEM of at least 3 experiments.
