The membrane-anchored PC1 tail activates STAT3. (A) PC1 expression constructs. Membrane-anchored constructs contain the extracytoplasmic domain and signal peptide (SP) of CD16 and transmembrane domain of CD7. (BD) Luciferase assays using HEK293T cells transfected with the STAT1/3 luciferase reporter and either control GFP or (B) wild-type PC1, noncleavable GPS-domain mutant PC1, or membrane-anchored PC1 tail or (C) FLM-PC1 truncation constructs or (D) pathogenic mutants in the FLM-PC1 context.
