Effect of a single concentration of S1P receptor-specific antagonist on MLC phosphorylation in the presence of increasing concentrations of S1P. Cells were treated with 1 U/ml thrombin (T) and S1P at concentrations of 10, 1, and 0.1 μM for 5 min and probed for MLC phosphorylation. S1P effects in the presence of antagonists were tested in additional cells pretreated with 10 μM of JTE-013 (S1P₂), W146 (S1P₁), or VPC23019 (S1P_1,3) for 30 min. SC (A) and TM (B) blots are representative of five different experiments using three cell strains for each cell type. β-Actin was used as a loading control.
