Mixing of mutant Cx50 but not wild-type Cx50 with Cx46 results in increased hemichannel opening at negative potentials. AD: membrane currents recorded in response to voltage-clamp steps from −80 mV to +30 mV in 10-mV increments from a holding potential of −80 mV. Equal amounts of Cx46 cRNA were injected into each oocyte. Oocytes were bathed in NaCl solution containing 0.7 mM [Ca2+]o and 1 mM [Mg2+]o. E: tail I-V relationships for Cx46 (■, n = 3), Cx46 + G46V (5:1, ○, n = 3), and Cx46 + G46V (1:2, ▵, n = 3). Initial amplitudes of tail currents were measured at − 80 mV after 20-s pulses to different potentials and plotted as a function of pulse potential. F: tail I-V relationships for Cx46 (■, n = 3), Cx46 + Cx50 (5:1, ○, n = 3), and Cx46 + Cx50 (1:5, ▼, n = 3). Initial amplitudes of tail currents were measured at −80 mV after 20-s pulses to different potentials and plotted as a function of pulse potential.
