FLuc reporter constructs in pcDNA3 vector bearing either wild type (left) or mutant (right) CARE in the HSR were transfected into HEK293T cells with CARE-binding miRNA candidates (miR-297, -299, -567, and -609), miR-control (Cont.), negative controls (miR-410 and -369), and with RLuc-expressing pRL-SV40 vector as internal control. Relative Luc levels were measured after 24 h and expressed as percentage of control. Results are expressed as mean±s.d., for n=3 independent experiments. An asterisk (*) indicates a significant difference, P<0.05, two-tailed t-test.
