Absence of Glc3Man5GlcNAc2-P-P-dolichol in Lec35 mutant cells. Lec35.1 cells (A and B) and Lec15.2 cells (Camp et al., 1993) (C and D) were labeled with 1 mCi/ml [2-3H]mannose (A and C) or [1-3H]galactose (B and D) for 15 min as described under MATERIALS AND METHODS. Lipid-linked oligosaccharides were extracted, and the labeled oligosaccharides were released by mild acid hydrolysis and analyzed by HPLC.
