TNF-α receptor-mediated signaling is essential to myogenic activation of p38. Primary myoblasts were isolated from 3- to 4-day-old wild-type (WT) or p55−/−p75−/− double-knockout (KO) mice. Differentiation was induced by replacing GM with DM. Myoblasts were collected at 48 h of differentiation. Activation of p38, Akt, ERK1/2, and JNK were analyzed as described in Fig. 2. Representative blots from 2 independent experiments are shown. Data are expressed as x-fold of the level at 0 h.
