Dose-dependent inhibition of COX-2 expression by fisetin, luteolin, and apigenin. HBMEC were serum-starved in the presence of various concentrations of fisetin, luteolin, and apigenin in combination with vehicle or 1 μM PMA for 18 hours. A) Lysates were isolated, electrophoresed via SDS-PAGE, and immunodetection of COX-2 and GAPDH performed as described in the Materials and Methods section. B) Scanning densitometry of COX-2 expression was only performed in PMA-treated cells since no COX-2 was detectable in vehicle-treated cells. Densitometric data of a representative blot out of three is shown.
