Raf-1 activation prevents cytochrome c/ATP-mediated activation of caspase 9. CCL39-ΔRaf-1:ER cells were deprived of serum for 9 hours in the presence or absence of 4-HT. The cells were lysed in hypotonic buffer and the cell lysates were incubated with 1 μM cytochrome c and 1 mM ATP for the indicated times. The cell lysates were analyzed by immunoblotting for caspase-9, phospho-, and total MAPK1,3.
