Inactivation of E(z) is insufficient to turn on terminal differentiation genes in a tTAF mutant background. (A) Immunoblot analysis showing the H3K27me3 band in nht testes and the drastic reduction of H3K27me3 in nht; E(z) double-mutant testes. H3 provided a loading control. (B) Quantitative RT-PCR analysis of dj, fzo and Mst87F transcript levels in nht; E(z), nht and wild-type (wt) testes, normalized to Rpl32 from the same sample. The nht; E(z), nht and wild-type flies were all raised to adulthood at permissive temperature and then shifted to 29°C (restrictive temperature) for 7 days. The relative level of each gene in the wild type was set to 1 to allow comparison. Error bars indicate s.d. from three independent biological replicates.
