Time-lapse images of two vesicles during domain collapse. The CL-containing GUVs are labelled with the NBD-PE (green) and Rh-PE (red) probes. The origin for the timescale for each vesicle is from the start of observation of the vesicle and not the time cyt c was added to the samples. The NBD-PE dye photobleaches during the time-lapse imaging: a green dotted line is drawn in the final image of vesicle A to highlight the location of the Lo domain. Vesicle A is monitored over a period of approximately 11 min and vesicle B is monitored over a period of approximately 2 min. Insets (i–iv) show magnified sections of vesicle A’s membrane where the budded structure within the Ld domains can be clearly resolved.
