(A.) Cells were incubated as described in Fig. 1 except that 1µM hyoscyamine (Hy) was added to some plates 60 min prior to addition of CCh or serum-free DMEM (Control, C) for 5 min. Results shown are the mean ± SEM of the increase in normalized HSP27 phosphorylation at Ser-82 from 3–4 experiments. (B.) Cells were preincubated for 1 hr in serum-free DMEM prior to addition of 1mM CCh or nicotine (Nic) or 10 −7 M bradykinin (BK) for 1 or 5 min. The insert shows a single immunoblot of phospho-(Ser-82)- and total HSP27. Results from 3–4 experiments were combined to obtain the mean ± SEM of the normalized HSP27 phosphorylation. ** p< 0.01 as compared to the control value.
