The loading of siRNAs is less sensitive to thermodynamic stability than that of shRNAs. (A) Wild-type or Ago2 KO MEF cells were cotransfected with psiCHECK reporter with four tandem mir30 target sites in the 3′ UTR and various synthetic siRNA duplexes. Dual-luciferase assays were performed at 24 h posttransfection, and the results are plotted as described previously. Error bars represent the SD from three independent experiments, each performed in triplicate. (B) The same experiment was performed with synthetic shRNAs, the sequences of which were exactly as illustrated in Fig. 2A.
