Assembly-induced change in tryptophan-induced quenching. The cys of Y222W/T151C, where the alpha carbons of W and C are 10 Å apart, was labeled with ATTO-655 or BODIPYaem and examined for quenching. (a) ATTO fluorescence in GDP was substantially quenched in the native FtsZ relative to urea-denatured FtsZ. Adding GTP to induce assembly reduced the quenching, indicated by increased fluorescence emission. (c) Quenching of BODIPYaem fluorescence in the Y222W/T151C mutant was very similar to that of ATTO. (d) BODIPYaem-labeled Y222W/T154C, where the W and C are only 7.2 Å apart, showed an insignificant fluorescence increase after adding GTP to induce the assembly.
