Kinetics of PNA1 binding to three target sequences containing a single mismatch. The PvuII-PvuII fragment of plasmid pSM-1, pSM-2 or pSM-3 (312 bp) was incubated at 42°C with γ-PNA in the presence of 35 mM Na+ at various times. The PNA concentration was 1.5 µM and the dsDNA concentration was 10 nM. (A) Plots of fraction of complexes as a function of time with targets SM-1 (squares), SM-2 (triangles) or SM-3 (circles).
