Cytokine (Il-2 and IFN-gamma) secretion in splenocytes from MMTV.f.huHER2(Fo5) transgenic mice immunized with PBS or anti-Id scFv69. Splenocytes were isolated on Day 42 post-immunization. Supernatants from antigen-stimulated cells were recovered 24 h (for IL-2) and 72 h (for IFN-gamma) after in vitro stimulation. IL-2 and IFN-gamma detection (ng/ml) were performed using an ELISA assay, with a standard curve obtained with recombinant IL-2 (detection limit 4 ng/ml) or recombinant IFN-gamma (detection limit 1 ng/ml). Results are the mean ± SD of triplicate wells and are representative of two independent experiments. Mfe23-Fc and scFv13R4 are two irrelevant antigens. Con A, Concanavalin A.
