Table 1.

Starch content and amylase activities in the leaf blades

Plant Starch content (% FW)
α-Amylase activity (U FW g−1)a β-Amylase activity (U FW g−1)b
End of the day End of the night
Wild type 1.98 ± 0.04 0.05 ± 0.01 0.96 ± 0.04 1.02 ± 0.01
isa3 2.09 ± 0.06 1.23 ± 0.03* 1.10 ± 0.01* 1.16 ± 0.01*

The values are the average for three replications (mean ± SE).

Significant differences from the wild type are marked with an asterisk (P < 0.01, t-test).

a One unit of α-amylase activity, Ceralpha Unit (Megazyme), is defined as the amount of enzyme, in the presence of excess α-glucosidase and glucoamylase, required to release 1 μmol of p-nitrophenol from BPNPG7 in 1 min under the defined assay conditions.

b One unit of β-amylase activity, Betamyl Unit (Megazyme), is defined as the amount of enzyme required, in the presence of excess β-glucosidase, to release 1 μmol of p-nitrophenol from PNPG5 in 1 min under the defined assay conditions.
