Substitution of the U51–U63 T-stem mismatch in tRNASer(CGA) with a G:C or A:U pair restores growth to a trm44tan1-Δ strain and prevents tRNASer(CGA) degradation. (A) Replacement of U51–U63 of tRNASer(CGA) with G:C or A:U restores growth of a trm44tan1-Δ strain at 37°C. Strains were grown and plated as in Figure 1B. (B) tRNASer(CGA) variants are resistant to degradation in a trm44tan1-Δ strain upon replacement of U51–U63 of tRNASer(CGA) with G:C or A:U. Strains were grown and treated as described in Figure 1C, and 3 μg of bulk RNA was analyzed by Northern blot. (C) Quantification of tRNASer(CGA) and tRNASer(UGA) levels 8 h after temperature shift. Levels of tRNASer species were normalized to tRNAPhe levels in the same sample, and then divided by the normalized levels of the same tRNA species in the wild-type parent strain (BY4741) prior to temperature shift.
